If you're relocating a plumbing vent, then move the cover you've got, and rivet a (properly caulked) circular aluminum patch over the former hole.

Not a
bad design, either. The amount of precipitation that would go down the vent stack is trivial unless you live in the Olympic peninsula or south Louisiana, and that rainfall would only end up in your holding tanks anyway. Your home vent stacks are open to the elements as well. There is no harm in leaving your new vent stack open the way the old one was.
