OSB floor, I assume? I have patched several holes in my trailer. I cut out the damaged area and fit a new piece. I slide material down through the hole to make a cleat on the underside of the floor to hold the new piece. Then glue and screws.
Fixing the leak is necessary. And easier to do before you patch the floor. If the ares is wet, I would cut it out and look for the source of the leak. If it is dry, then maybe you could just fill it in with the rotten wood epoxy.
