Sub Floor Patch

Hello - I am replacing an area of rotten floor 7" x 48" at the rear of my 1964 Bambi. I have not removed the inner skins (there would be 2 to remove if I have to) as I was hoping to glue it in place as far under the inner skin as I can get it. I do not think I will be able to get it into the "C" channel. This piece of repaired flooring will be under the bathroom vanity unit and so hidden from view or being stood on. After repairing I was thinking of gluing 1/4 inch plywood over all of my sub floor to give a good even base for maybe vinyl or whatever flooring I choose.
My questions are:-
Should I really remove the inner skins to fix it under the "C" channel properly?
Secondly the best way to affix this piece bearing in mind it will be hidden?
And lastly is the 1/4 plywood a good idea or a waste of time and money?
